Description 4-1BB (CD137) is a co-stimulatory molecule belonging to the tumor necrosis factor receptor (TNFR) superfamily. It is primarily expressed on activated T cells, natural killer (NK) cells, and other immune cells. Its ligand, 4-1BBL (CD137L), is mainly expressed on antigen-presenting cells (APCs) such as dendritic cells and macrophages. 4-1BB plays a critical role in the immune system, particularly in regulating T cell activation, proliferation, and survival. Urelumab binds to 4-1BB and activates the co-stimulatory signaling of T cells and NK cells, thereby enhancing anti-tumor immune responses. As a representative 4-1BB agonist, Urelumab has shown significant potential in boosting anti-tumor immune responses, despite being limited by toxicity issues in early studies.
